RESUMO

To explore contaminant concerns as a result of anthropogenic disturbance of the river system, this study provided the first extensive investigation of the contamination profiles, possible driving factors, and ecological risks of 40 target compounds including pharmaceuticals and personal care products (PPCPs), neonicotinoid pesticides (NNIs), polybrominated diphenyl ethers (PBDEs), and polychlorinated biphenyls (PCBs) in sediments of the whole Yangtze River (the world's third longest river). Among these target compounds, PPCPs were the dominant contaminants with a total concentration (∑15PPCPs) of 2.13-14.99 ng/g, followed by ∑7PCBs (

Purpose: To establish nomograms integrating serum lactate levels and traditional risk factors for predicting diabetic kidney disease (DKD) in type 2 diabetes mellitus (T2DM) patients. Patients and methods: A total of 570 T2DM patients and 100 healthy subjects were enrolled. T2DM patients were categorized into normal and high lactate groups. Univariate and multivariate logistic regression analyses were employed to identify independent predictors for DKD. Then, nomograms for predicting DKD were established, and the model performance was evaluated using the area under the receiver operating characteristic curve (AUC), calibration, and decision curve analysis (DCA). Results: T2DM patients exhibited higher lactate levels compared to those in healthy subjects. Glucose, platelet, uric acid, creatinine, and hypertension were independent factors for DKD in T2DM patients with normal lactate levels, while diabetes duration, creatinine, total cholesterol, and hypertension were indicators in high lactate levels group (P<0.05). The AUC values were 0.834 (95% CI, 0.776 to 0.891) and 0.741 (95% CI, 0.688 to 0.795) for nomograms in both normal lactate and high lactate groups, respectively. The calibration curve demonstrated excellent agreement of fit. Furthermore, the DCA revealed that the threshold probability and highest Net Yield were 17-99% and 0.36, and 24-99% and 0.24 for the models in normal lactate and high lactate groups, respectively. Conclusion: The serum lactate level-based nomogram models, combined with traditional risk factors, offer an effective tool for predicting DKD probability in T2DM patients. This approach holds promise for early risk assessment and tailored intervention strategies.

This study investigated the pollution of perfluorinated compounds (PFCs) in sediments from the main stream of the Yangtze River, the longest river in Asia. Totally, 13 of 15 PFASs were detected in the sediments and the total concentrations ranged from 0.058 ng/g to 0.89 ng/g dry weight (dw), with dominant contaminants by perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA). Concentrations of PFASs in the downstream were higher than those of upstream and midstream. Four main sources were analysed using the Unmix model, textile treatments and food packaging dominantly accounted for approximately half of the total sources, followed by metal electroplating (26.8%), fluoropolymer products (16.3%) and fluororesin coatings (7.4%). Total organic carbon (TOC), total nitrogen (TN) and grain size had significant correlation with the concentration of PFASs in sediments, indicating that the physical and chemical parameters could directly affect the adsorption process of PFASs. In addition, anthropogenic factors such as urbanization rate and per capita GDP also had a direct impact on the distribution of PFASs. Environmental risk assessment showed that PFOS posed low to medium risks to the Yangtze River, indicating that sustained attentions were needed.

Antibiotic resistance is an escalating global health concern, exacerbated by the pervasive presence of antibiotic resistance genes (ARGs) in natural environments. The Yangtze River, the world's third-longest river, traversing areas with intense human activities, presents a unique ecosystem for studying the impact of these genes on human health. Here, we explored ARGs in the Yangtze River, examining 204 samples from six distinct habitats of approximately 6000 km of the river, including free-living and particle-associated settings, surface and bottom sediments, and surface and bottom bank soils. Employing shotgun sequencing, we generated an average of 13.69 Gb reads per sample. Our findings revealed a significantly higher abundance and diversity of ARGs in water-borne bacteria compared to other habitats. A notable pattern of resistome coalescence was observed within similar habitat types. In addition, we developed a framework for ranking the risk of ARG and a corresponding method for calculating the risk index. Applying them, we identified water-borne bacteria as the highest contributors to health risks, and noted an increase in ARG risks in particle-associated bacteria correlating with heightened anthropogenic activities. Further analysis using a weighted ARG risk index pinpointed the Chengdu-Chongqing and Yangtze River Delta urban agglomerations as regions of elevated health risk. These insights provide a critical new perspective on ARG health risk assessment, highlighting the urgent need for strategies to mitigate the impact of ARGs on human health and to preserve the ecological and economic sustainability of the Yangtze River for future human use.

PURPOSE: Numerous clinical studies have explored sodium-glucose cotransporter 2 inhibitor (SGLT2i) in patients with chronic heart failure (CHF), with or without type 2 diabetes mellitus (T2DM), and SGLT2i were proved to significantly reduce CHF hospitalization, cardiovascular death, cardiovascular mortality, all-cause mortality and myocardial infarction in patients with or without T2DM. However, only a limited few have investigated the effects of SGLT-2i on HF disease-specific health status and cardiac function. This meta-analysis aims to assess the effects of SGLT2i on disease-specific health status and cardiac function in CHF patients. METHODS: A comprehensive search was conducted of trials by searching in PubMed, EMBASE, CENTRAL, Scopus, and Web of Science, and two Chinese databases (CNKI and Wanfang), Clinical Trials ( http://www. CLINICALTRIALS: gov ) were also searched. RESULTS: A total of 18 randomized controlled trials (RCTs) involving 23,953 participants were included in the meta-analysis. The effects of SGLT2 inhibitors were compared with control or placebo groups in CHF with or without T2DM. The SGLT2 inhibitors group exhibited a significant reduction in pro b-type natriuretic peptide (NT-proBNP) levels by 136.03 pg/ml (95% confidence interval [CI]: -253.36, - 18.70; P = 0.02). Additionally, a greater proportion of patients in the SGLT2 inhibitors group showed a ≥ 20% decrease in NT-proBNP (RR = 1.45, 95% CI [0.92, 2.29], p = 0.072). However, no statistically significant difference was observed for the effects on B-type natriuretic peptide (BNP). The use of SGLT-2 inhibitors led to a noteworthy improvement in LVEF by 2.79% (95% CI [0.18, 5.39];P = 0.036). In terms of health status, as assessed by the Kansas City Cardiomyopathy Questionnaire (KCCQ) and 6-minute walk distance, SGLT2 inhibitors led to a significant improvement in KCCQ clinical summary (KCCQ-CS) score (WMD = 1.7, 95% CI [1.67, 1.73], P < 0.00001), KCCQ overall summary (KCCQ-OS) score (WMD = 1.73, 95% CI [0.94, 2.52], P < 0.00001), and KCCQ total symptom (KCCQ-TS) score (WMD = 2.88, 95% CI [1.7, 4.06], P < 0.00001). Furthermore, the occurrence of KCCQ-CS and KCCQ-OS score increases ≥ 5 points had relative risks (RR) of 1.25 (95% CI [1.11, 1.42], P < 0.00001) and 1.15 (95% CI [1.09, 1.22], P < 0.00001), respectively. Overall, SGLT2 inhibitors increased the 6-minute walk distance by 23.98 m (95% CI [8.34, 39.62]; P = 0.003) compared to control/placebo from baseline. CONCLUSIONS: The SGLT2 inhibitors treatment offers an effective strategy for improving NT-proBNP levels, Kansas City Cardiomyopathy Questionnaire scores and 6-minute walk distance in CHF with or without T2DM. These findings indicate that SGLT2i improve cardiac function and health status in CHF with or without T2DM, and provide valuable guidance for clinicians making treatment decisions for patients with CHF.

As important freshwater ecosystems, the occurrence and distribution of antibiotic resistance genes (ARGs) in rivers are relevant to public health. However, studies investigating ARGs of different environmental media in river ecosystems are limited. In this study, we analyzed the ARGs of microbes in free-living setting, particle-associated setting, sediment and bank soil of the Yangtze River using metagenomics. Twenty-six ARGs were found in all samples regardless of media (core resistome) with a diversity of 8.6 %-34.7 %, accounting for 22.7 %-89.2 % of the relative abundance of the overall ARGs. The core resistome of the Yangtze River was dominated by multidrug resistance genes consisting mainly of efflux pumps and bacitracin resistance genes. The rare resistome was dominated by multidrug, sulfonamide, and aminoglycoside resistance genes. The core resistome was more prevalent in chromosomes, implying that these ARGs with low diversity and high relative abundance may be intrinsic to microbes in the Yangtze River. The rare resistome was more prevalent in plasmids, suggesting these ARGs with high diversity and low relative abundance were acquired under environmental stresses and had transfer potential. Additionally, we found that core and rare resistome were mainly carried by specific bacteria. Noteworthily, twenty-two ARGs of high clinical concern were identified in rare resistome, especially aac(6')-I, sul1, and tetM, which were plasmid-borne and hosted by clinically relevant pathogens. Both core and rare resistome hosts showed the highest niche breadths in particle-associated setting compared to other media, and particle-associated setting could provide more stable and ideal conditions for resistome hosts to survive. This study elucidated the genetic locations of ARGs and the community assembly mechanisms of ARG hosts in freshwater environments.

A 49-year-old man presented with a 2-year history of weakness and sensory disturbances in the bilateral lower extremities, vesicorectal dysfunction, and progressive gait disturbances. Brain MRI revealed multiple ischemic and hemorrhagic cortical/subcortical lesions with patchy enhancement involving the frontal and parietal lobes, suggesting the possibility of distal perforating arteries injury. Spine MRI revealed lesions of the cervical and thoracic spinal cord with associated enhancement. The diagnosis of malignant atrophic papulosis (Degos disease) with central nervous system involvement was prompted by the characteristic skin lesions.

Phthalate esters (PAEs) are the most ubiquitous and highly used plasticizers in plastic products globally, yet studies on the spatial variation, risks, and their correlation with microplastics (MPs) are limited, particularly throughout the Yangtze River (the largest river in China/Asia). Therefore, this study investigated for the first time the PAEs pollution characteristics throughout the Yangtze River sediments, studied the environmental factors linked to the distribution of PAEs, and explored their potential as chemical indicators for interpreting pollution patterns of MPs. Totally 14 out of 16 PAEs were detected in sediments, with total concentrations ranging from 84.67 ng/g to 274.0 ng/g (mean: 163.5 ng/g), dominated by Bis(2-ethylhexyl) phthalate (DEHP), Di-n-butyl phthalate (DBP), and Di-isobutyl phthalate (DIBP), with contributions of 38.9 %, 31.8 %, and 20.8 %, respectively. Spatial distribution of PAEs did not indicate significant differences, which may be related to anthropogenic activities (i.e., emission intensity), runoff, and sediment physicochemical properties (i.e., TOC and TN), with TOC and TN being potential predictors of PAEs. The quantitative relationships (p < 0.001) between DEHP/∑16PAEs ratio and MPs (both individual and total MPs) were found in sediments, which suggested that DEHP could be potentially used as an indicator for MPs. DEHP, DIBP, and DBP posed high risks, accounting for 100 %, 68.4 %, and 10.5 % of the monitoring sites, respectively. Further work is necessary to better understand the relationship between DEHP/∑16PAEs and MPs in the environment and to take corresponding management and control measures for these pollutants.

Although NG2 is known to be selectively expressed in oligodendrocyte precursor cells (OPCs) for many years, its expressional regulation and functional involvement in oligodendrocyte differentiation have remained elusive. Here, we report that the surface-bound NG2 proteoglycan can physically bind to PDGF-AA and enhances PDGF receptor alpha (PDGFRα) activation of downstream signaling. During differentiation stage, NG2 protein is cleaved by A disintegrin and metalloproteinase with thrombospondin motifs type 4 (Adamts4), which is highly upregulated in differentiating OPCs but gradually downregulated in mature myelinating oligodendrocytes. Genetic ablation of Adamts4 gene impedes NG2 proteolysis, leading to elevated PDGFRα signaling but impaired oligodendrocyte differentiation and axonal myelination in both sexes of mice. Moreover, Adamts4 deficiency also lessens myelin repair in adult brain tissue following Lysophosphatidylcholine-induced demyelination. Thus, Adamts4 could be a potential therapeutic target for enhancing oligodendrocyte differentiation and axonal remyelination in demyelinating diseases.SIGNIFICANCE STATEMENT NG2 is selectively expressed in OPCs and downregulated during differentiation stage. To date, the molecular mechanism underlying the progressive removal of NG2 surface proteoglycan in differentiating OPCs has been unknown. In this study, we demonstrate that ADAMTS4 released by differentiating OPCs cleaves surface NG2 proteoglycan, attenuates PDGFRα signaling, and accelerates oligodendrocyte differentiation. In addition, our study also suggests ADAMTS4 as a potential therapeutic target for promoting myelin recovery in demyelinating diseases.

AIM: Early mobilization (EM) improves critical illness survivors' clinical outcomes. This study examines ICU quasi-specialty nurses' EM-related knowledge, attitudes and practices (KAP). DESIGN: This cross-sectional study was conducted at a provincial ICU specialty nurse training site from 2019 to 2021. KAP data were collected using a self-report questionnaire. METHODS: Participants were registered nurses enrolled in the training programme; 485 nurses from 188 hospitals completed the questionnaire. RESULTS: Of the participants, 37.7% had EM training, and 30.7% reported their wards performed EM. Median (IQR) scores for knowledge, attitudes and practices were 14.0 (4.0), 47.0 (9.0) and 37.0 (8.0), belonging to medium, high and medium levels. Scores were lowest for EM scope, implementation indicators and out-of-bed mobilization. There were significant deviations in KAP scores amongst nurses at different levels of hospitals with EM training and whose departments performed EM. Significant positive correlations between knowledge and practice, and attitude and practice were identified.

The retina is one of the most metabolically active tissues in the body. The dysfunction of oxygen kinetics in the retina is closely related to the disease and has important clinical value. Dynamic imaging and comprehensive analyses of oxygen kinetics in the retina depend on the fusion of structural and functional imaging and high spatiotemporal resolution. But it's currently not clinically available, particularly via a single imaging device. Therefore, this work aims to develop a retinal oxygen kinetics imaging and analysis (ROKIA) technology by integrating dual-wavelength imaging with laser speckle contrast imaging modalities, which achieves structural and functional analysis with high spatial resolution and dynamic measurement, taking both external and lumen vessel diameters into account. The ROKIA systematically evaluated eight vascular metrics, four blood flow metrics, and fifteen oxygenation metrics. The single device scheme overcomes the incompatibility of optical design, harmonizes the field of view and resolution of different modalities, and reduces the difficulty of registration and image processing algorithms. More importantly, many of the metrics (such as oxygen delivery, oxygen metabolism, vessel wall thickness, etc.) derived from the fusion of structural and functional information, are unique to ROKIA. The oxygen kinetic analysis technology proposed in this paper, to our knowledge, is the first demonstration of the vascular metrics, blood flow metrics, and oxygenation metrics via a single system, which will potentially become a powerful tool for disease diagnosis and clinical research.

BACKGROUND: Riverine ecosystems are one of the most important reservoirs of antibiotic resistance genes (ARGs) in the environment, but the occurrence and controlling factors of ARG distribution in different habitats of riverine ecosystems remain poorly understood. In this study, a metagenomic approach was used to characterize ARG types and their abundance in different habitats (rhizosphere soil, surface bulk soil, bottom bulk soil, and sediment) of riverine ecosystems in eastern China. Sampling sites were located along different rivers of eastern China, which are geographically isolated. Differences in bacterial communities, mobile genetic elements (MGEs), pattern and intensity of human activities, climate, and other environmental factors at the sampling sites and habitats were expected to affect ARG occurrence. RESULTS: ARGs were observed with high variations in diversity (44-206 subtypes) and abundance (6.85-105.68 ×/Gb). There were significant south-north differences in ARG occurrence in the same habitat, except for surface bulk soil. And the significant difference was found in ARGs among four southern habitats. South-north differences in ARGs of the same habitat were mainly attributed to the combination of different occurrence frequencies and habitat selections of ARGs. Differences in ARG profiles among the four habitats in the south and the north were both mainly attributed to the different occurrence frequencies of ARGs. Bacterial communities and MGEs (Mobile genetic elements) could account for the observed variance in the resistome of riverine ecosystems across eastern China. The co-occurrences of specific ARGs with bacterial communities and MGEs were more frequent at the northern sampling sites than in the south, and co-occurrence patterns (i.e. ARGs and bacterial communities or ARGs and MGEs) varied between the habitats. Moreover, building land in all habitats, except bulk soils, showed significant positive correlations with ARG abundance. CONCLUSION: This study reveals a high variance in the resistome of riverine ecosystems in eastern China and its controlling factors. We appeal to the importance of assessment of ARGs in the riverine ecosystem and the need for future prevention and intervention of ARG spread.

This study conducted the first extensive and comprehensive investigation of the whole-scale sedimentary antibiotic concentration, possible drivers, environmental fate, and potential ecological risks in the Yangtze River. Totally, 20 antibiotics were detected in the sediments. Results revealed that the order of antibiotic abundance in sediment was fluoroquinolones > tetracyclines > macrolides > sulfonamides > amphenicols. The total antibiotic concentrations were 0.10-134.4 ng/g (mean: 11.88 ng/g). Of these, fluoroquinolones and tetracyclines were the two dominant antibiotic categories. The dominant occurrence of fluoroquinolones and tetracyclines in sediments suggested that the distribution coefficient (Kd) was one of the important factors to determine their fate. Correlation analysis demonstrated that antibiotic contamination was largely influenced by the local scale of animal husbandry, and the positive correlation between antibiotics and heavy metals was likely driven by their common source of contamination and the complexation. Environmental risk assessment showed that tetracycline and chlortetracycline exhibited potential risks from medium to high in the Yangtze River, although most of the compounds posed minimal and low risks. This work provided a valuable large-scale data set across the whole Yangtze River and revealed the contamination profile of antibiotics. Mitigation and management measures to reduce antibiotic inputs are needed for the Yangtze River basin.

Plastic surfaces in the environment are a comparatively new niche for microbial colonization, also known as the "plastisphere". However, our understanding of the core and occasional bacterial taxa in the plastisphere is limited. Here, environmental plastic, water, and sediment samples were collected from 10 sites in a plateau river (Lhasa River, China) in September of 2019. The composition and spatial turnover of core and occasional bacterial taxa in the plastisphere were revealed via 16S rRNA gene sequencing and compared with water and sediment. The results indicated that deterministic processes dominated the habitat specialization that shaped the formation of core and occasional taxa in the plastisphere, water, and sediment of the Lhasa River because the decline in zeta diversity in the plastisphere, water, and sediment was more fitted to a power-law form rather than an exponential form. Proteobacteria (65.9%), Bacteroidetes (16.0%), and Cyanobacteria (11.7%) dominated the plastic core taxa. Core taxa rather than occasional taxa in the plastisphere had a lower (21.7%) proportion of OTUs and a higher (81.7%) proportion of average relative abundance than water and sediment, which were dominant in plastic bacterial communities. The spatial turnover of core and occasional bacterial taxa in the plastisphere was governed by abiotic as well as biotic factors. Specifically, the spatial turnover of core taxa in the plastisphere with high connectivity but low functional redundancy was easily affected by geographical distance, altitude, and heavy metals. Furthermore, strong drug resistance was found in the spatially persistent core taxa in the plastisphere. This study provides empirical support for the spatial turnover (species variation) and potential ecological mechanisms of bacterial communities in the plastisphere from river ecosystems.

This paper reports Pb isotopes in different fractions following the three step BCR and 1 M HCl extractions on river sediments from lower reaches of the Xiangjiang river in China, and highlights the importance of Pb isotopes in heavy metal contamination assessment. Lead concentrations and Pb isotopes in bulk sediments and sediment fractions (leachates and residues) from the river were analysed using ICP-MS techniques. Results showed that sediments were highly enriched with Pb with enrichment factors >5.5, while Pb in sediments was dominated by reducible and residual Pb fractions, residing mainly in Fe-oxide and silicate minerals. Pb isotopes in sediments was characterized by radiogenic Pb produced from the decay of uranium and thorium with 206Pb/207Pb ratios of 1.1744 for less radiogenic Pb and 1.1816 for more radiogenic Pb. The leachates and residues from BCR extraction generally had similar Pb isotope compositions, of which the 206Pb/207Pb ratios were 1.1798 ± 0.002 and 1.1844 ± 0.008 respectively. Differentiation of Pb isotopes between BCR leachates and residues was insignificant. However, differentiation between leachates and residues using 1 M HCl extraction was significant, as shown by average 206Pb/207Pb ratios of 1.1746 ± 0.005 and 1.1858 ± 0.008 for leachates and residues respectively. Pb isotopic tracing suggests that Pb in sediments from Zhuzhou section arose from the mixing of anthropogenic Pb from coal combustion (39%) and mining-smelting for Pb-Zn ores (58%); while Pb in sediments from Xiangtan, Changsha and Xiangyin sections arose from the mixing of anthropogenic Pb from mining-smelting for Pb-Zn ores (54%), and lithologically inherited Pb from granite weathering (35%) with a small amount of contribution from coal combustion (10%). The present study suggests that the BCR extraction scheme was not appropriate for ecological risk assessment of heavy metal contamination in mining-impacted (ore-Pb dominated) river sediments.

Systemic vascular impairment is the most common complication of diabetes. Advanced glycation end products (AGEs) can exacerbate diabetes-related vascular damage by affecting the intima and media through a variety of mechanisms. In the study, we demonstrated that AGEs and their membrane receptor RAGE could induce the differentiation of EPCs into osteoblasts under certain circumstances, thereby promoting accelerated atherosclerosis. Differentiation into osteoblasts was confirmed by positive staining for DiI-acetylated fluorescently labeled low-density lipoprotein and FITC-conjugated Ulex europaeus agglutinin. During differentiation, expression of receptor for AGE (RAGE) was significantly upregulated. This upregulation was attenuated by transfection with RAGE-targeting small interfering (si)RNA. siRNA-mediated knockdown of RAGE expression significantly inhibited the upregulation of AGE-induced calcification-related proteins, such as runt-related transcription factor 2 (RUNX2) and osteoprotegerin (OPG). Additional experiments showed that AGE induction of EPCs significantly induced ERK, p38MAPK, and JNK activation. The AGE-induced upregulation of osteoblast proteins (RUNX2 and OPG) was suppressed by treatment with a p38MAPK inhibitor (SB203580) or JNK inhibitor (SP600125), but not by treatment with an ERK inhibitor (PD98059), which indicated that AGE-induced osteoblast differentiation from EPCs may be mediated by p38MAPK and JNK signaling, but not by ERK signaling. These data suggested that AGEs may bind to RAGE on the EPC membrane to trigger differentiation into osteoblasts. The underlying mechanism appears to involve the p38MAPK and JNK1/2 pathways, but not the ERK1/2 pathway.

PURPOSE: To compare the plasma immune globulin and complement levels in different sorts of glaucoma to assess its association. METHODS: Cohort study. Plasma samples were collected from 226 normal controls and 236 glaucoma patients included 92 with acute primary angle-closure glaucoma (APACG), 76 with chronic primary angle-closure glaucoma (CPACG), 68 with primary open-angle glaucoma (POAG). 163 glaucoma patients with high intraocular pressure(IOP) was classified as a subgroup. Six indexes were analyzed: C3, C4, CH50, IgA, IgG, and IgM. RESULTS: There was a significant difference in C4, IgA, IgG, and IgM between patients with high IOP and normal controls groups(P < 0.01). TheC4 (0.27 ± 0.69 g/L), IgA (3.11 ± 1.04 g/L), IgG (12.07 ± 1.92 g/L) and IgM (1.17 ± 0.49 g/L) were higher(P < 0.05) in the high IOP groups compared with the normal controls (C4:0.25 ± 0.07 g/L, IgA: 2.54 ± 1.07 g/L, IgG: 11.21 ± 2.22 g/L, IgM:0.99 ± 0.41 g/L). There was a significant difference in age, C3, IgA, IgG, and IgM among the normal control, APACG, CPACG, and POAG. The C3 level in the APACG group (1.14 ± 0.16 g/L) was higher than normal group (1.06 ± 0.18 g/L). The IgA level was lower(P < 0.05) in the normal group (2.54 ± 1.07 g/L) compared with APACG (3.16 ± 0.97 g/L) and POAG group (2.91 ± 1.11 g/L). The IgG level of normal controls (11.21 ± 2.22 g/L) was lower (P < 0.05) than APACG group (12.15 ± 2.02 g/L) and CPACG group (11.95 ± 2.28 g/L). The IgM level was lower(P < 0.05) in the normal group (0.99 ± 0.41 g/L) compared with the APACG (1.24 ± 0.66 g/L) and CPACG group (1.17 ± 0.45 g/L). CONCLUSIONS: The levels of the plasma expression of C3, IgA, IgG, IgM may be useful for discriminating the glaucoma patients and for assessing the progress of glaucoma in different sorts.

OBJECTIVES: Significance of long noncoding RNAs in pancreatic cancer (PC) progression has been documented. Here, we identified a novel long noncoding RNA MIR600HG in PC and its underlying mechanism during PC progression. METHODS: Through bioinformatics analysis, we selected MIR600HG, microRNA-125a-5p (miR-125a-5p), and mitochondrial tumor suppressor 1 (MTUS1) as objects with their expression patterns assayed in the collected PC tissues and PC cells. Pancreatic cancer cells were manipulated with ectopic expression and deficiency of MIR600HG, miR-125a-5p, and/or MTUS1 for assaying cell biological processes in vitro and tumorigenesis in vivo. RESULTS: MIR600HG and MTUS1 levels were downregulated and miR-125a-5p was upregulated in PC tissues and cells. MIR600HG could bind to miR-125a-5p, while miR-125a-5p negatively targeted MTUS1. MIR600HG resulted in suppression in malignant properties of PCs. All these changes could be reversed by miR-125a-5p elevation. In addition, miR-125a-5p targeted MTUS1 to activate the extracellular regulated protein kinases signaling pathway. In vivo experiment also verified the inhibitory role of MIR600HG in PC. CONCLUSIONS: Taken together, MIR600HG acts as an inhibitor for PC progression by upregulating miR-125a-5p-mediated MTUS1 through extracellular regulated protein kinases pathway.
